背景介绍(Background):
The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It encodes a protein which shows 96% amino acid identity to mouse Wnt3A protein, and 84% to human WNT3 protein, another WNT gene product. This gene is clustered with WNT14 gene, another family member, in chromosome 1q42 region. [provided by RefSeq, Jul 2008]
Wingless/Int (Wnt)-3a, one of the Wnt family members that has been regarded as an activator of the canonical Wnt signaling pathway, is expressed in the dorsal midline region responsible for developing spinal cord, forming a dorsal-toventral concentration gradient. During vertebrate embryogenesis, Wnt-3a participates in multiple cellular functions, including self-renewal, proliferation, differentiation, and motility. Wnt-3a was found to significantly activate the autophagy activity of neurons, inhibit the apoptotic activity of nerve cells, promote regeneration of axons, reduce the loss of motor neurons in the anterior horn of the spinal cord, and promote the repair of injured spinal cord tissue after spinal cord injury, providing a good microenvironment for the recovery of nerve function.
Wnt signaling regulates several biological processes, including brain development and stem cell proliferation, and has been implicated in a whole spectrum of diseases from cancer to neurodegeneration; Wnt-3a plays an important role in these processes. The Wnt signaling pathway can be divided into canonical and noncanonical pathways. The canonical pathway is also called the Wnt/β-catenin pathway. The canonical Wnt pathway begins with the binding of a Wnt ligand to its receptor Frizzled (Fz) and co-receptor lipoprotein receptor-related protein (LRP5/6), which then activates the scaffold protein Disheveled (Dvl) and causes dissociation of the “destruction complex” formed with casein kinase 1 (CK1), glycogen synthase kinase-3β (GSK-3β), and the scaffolding proteins adenomatous polyposis coli (APC), Axin1, and Axin2. The dissociation of the “destruction complex” causes the cytoplasmic accumulation of β-catenin and its translocation into the nucleus to interact with T cell–specific transcription factor (TCF) and lymphoid enhancer-binding factor (LEF), which regulate the expression of Wnt target genes.
