背景介绍(Background):
This protein binds to the hepatocyte growth factor receptor to regulate cell growth, cell motility and morphogenesis in numerous cell and tissue types. Alternative splicing results in multiple transcript variants, at least one of which encodes a preproprotein that is proteolytically processed to generate alpha and beta chains, which form the mature heterodimer. This protein is secreted by mesenchymal cells and acts as a multi-functional cytokine on cells of mainly epithelial origin. This protein also plays a role in angiogenesis, tumorogenesis, and tissue regeneration. Although the encoded protein is a member of the peptidase S1 family of serine proteases, it lacks peptidase activity. Mutations in this gene are associated with nonsyndromic hearing loss. Hepatocyte growth factor (HGF) was originally observed in rat serum and then characterized in human plasma. It is encoded by the human HGF gene (with a full-length sequence of around 71 kDa; https://www. ncbi.nlm.nih.gov/gene) It belongs to the peptidase S1 family, the plasminogen subfamily, and contains 6 domains (an N-terminal (N) domain and kringle 1-4 domains constitute the α-chain, and a serine protease homology (SPH) domain constitutes the β-chain) with 728 amino acids (https://www.uniprot. org/). HGF is secreted by a broad spectrum of tissues and cell types (predominantly stromal cells) and acts as a trigger for various cellular processes by binding its receptor c-MET through low- (in the SPH domain) or high-affinity binding sites (in the N and kringle 1 domains). Binding of HGF to MET induces conformational changes that result in receptor dimerization and trans-phosphorylation of tyrosine residues in the catalytic domain of MET and phosphorylation of tyrosine residues in the carboxyl-terminal tail. The phosphorylated tyrosine residues create docking sites for several adaptor molecules and kinase substrates as indicated. MET activation leads to subsequent activation of signaling pathways that include MAPK, PI3K/AKT and STAT3, which mediate MET-dependent cell proliferation, survival, migration and invasion.
