The antigenicity of the 15-kDa lipoprotein (Tpp15 or TpN15) of Treponema pallidum is modified with lipids.The native and the recombinant 15kD immunogens are amphiphilic proteins that appear to be located within the cytoplasmic membranes of their respective organisms. The lipoprolein structure of this potent immunogen is particularly significant in view of findings that the prototype bacterial lipoprotein, the murein lipoprotein of E.coli, and lipopeptides derived from its N-terminus, have unique B- and T-cell immunostimulatory properties. The Lipoprotein nature of the 15kD immunogen may have a direct bearing on the role of this molecule in syphilis immunopathogenesis.
